Potato extract with radiotherapy decrease colorectal cancer cells proliferation by increasing apoptosis

目的

探讨马铃薯提取物联合放疗对结直肠癌细胞增殖水平的影响及作用机制。

方法

采用人结直肠癌细胞株LS-174T,给予不同剂量X射线照射联合不同浓度马铃薯提取物作用,通过MTS法检测各组细胞增殖水平,Western blot法检测Bcl-2、Caspase-3在各组细胞中蛋白水平表达的情况,Annexin V-FITC/PI双染流式细胞术检测各组细胞凋亡情况。

结果

LS-174T细胞接受马铃薯提取物单独作用时,当浓度达到25 μL/mL后,对于细胞增殖的抑制作用逐渐增加,特别是在浓度为50 μL/mL作用72小时后对细胞增殖的抑制作用最显著(t=28.6,P=0.001)。马铃薯提取物联合放疗后,给予相同剂量时,马铃薯提取物浓度达到25 μL/mL时,细胞的增殖能力明显被抑制。放疗8 Gy联合马铃薯提取物50 μL/mL作用LS-174T细胞72 h后,细胞增殖抑制达到最高(t=12.1,P=0.007)。马铃薯提取物联合放疗使得LS-174T细胞的Bcl-2蛋白表达降低,Caspase-3蛋白呈高表达,流式细胞仪检测发现马铃薯提取物50 μL/mL联合放疗8 Gy能够促进细胞凋亡(t=18.3,P=0.003)。

结论

马铃薯提取物联合放疗能够显著抑制结直肠癌细胞的增殖,其作用机制可能是通过介导细胞凋亡发生,为结直肠癌放疗增敏剂的研究提供新的思路。

Objective

To investigate the effect of potato extract with radiotherapy on the proliferation of colorectal cancer cells and its mechanism.

Methods

Human colorectal cancer cell line LS-174T was used to give different doses of X-ray irradiation and different concentrations of potato extracts. MTS method was used to detect cell proliferation and tumor inhibition rate in each group, Western blot method to detect Bcl-2, Caspase-3 The expression of protein in each group of cells, Annexin V-FITC/PI double staining flow cytometry was used to detect cell apoptosis in each group.

Results

When LS-174T cells were treated with potato extract alone, which the concentration reached 25 μL/mL, the inhibitory effect on cell proliferation gradually increased. Especially, the inhibition of cell proliferation is most obvious with potato extract 50 μL/mL incubated for 72 hours (t=28.6, P=0.001). When the concentration of potato extract reaches 25 μL/mL combined with radiotherapy, the cell proliferation ability is obviously inhibited. After radiotherapy 8 Gy combined with 50 μL/mL of potato extract was treated on LS-174T cells for 72 h, the cell proliferation inhibited (t=12.1, P=0.007). Potato extract combined with radiotherapy reduced the expression of Bcl-2 protein in LS-174T cells, and the expression of Caspase-3 protein was high. Flow cytometry test showed that 50 μL/mL of potato extract combined with radiotherapy 8 Gy can promote cell apoptosis (t=18.3, P=0.003).

Conclusion

Potato extract combined with radiotherapy can significantly inhibit the proliferation of colorectal cancer cells, and its mechanism of action may be through mediating cell apoptosis, providing a new idea for the study of colorectal cancer radiosensitizers.
