目的

探讨长链非编码RNA小核仁RNA宿主基因17(SNHG17)在结直肠癌中的作用及其分子机制。

方法

采用实时荧光定量PCR(qRT-PCR)技术检测SNHG17在结肠癌细胞系和正常结肠上皮细胞中的表达;通过转染SNHG17质粒或SNHG17-shRNA,过表达或沉默SNHG17,检测其对结直肠癌细胞株迁移及侵袭能力的影响。

结果

SNHG17在结直肠癌组织和细胞系中表达增加,SNHG17的沉默能抑制结直肠癌细胞迁移和侵袭;SNHG17的上调促进结直肠癌细胞迁移和侵袭。通过检测蛋白水平发现SNHG17促进了结直肠癌细胞的上皮间质转化。

结论

LncRNA SNHG17在结直肠癌中表达较正常结肠组织增加,通过诱导上皮间质转化促进结直肠癌细胞的迁移和侵袭。

Objective

To investigate the role of long non-coding RNA small nucleolar RNA host gene 17 (SNHG17) and its molecular mechanism in colorectal cancer.

Methods

Quantitative Realtime PCR (qRT-PCR) was used to detect the expression of SNHG17 in colon cancer cell lines and normal colon epithelial cells; SNHG17 was overexpressed or silenced by transfection with SNHG17 plasmid or SNHG17-shRNA, to detect its effect on the migration and invasion of colorectal cancer cell lines.

Results

SNHG17 was up-regulated in colorectal cancer tissues and cell lines. The silencing of SNHG17 could inhibit colorectal cancer cell migration and invasion while the up-regulation of SNHG17 promoted colorectal cancer cell migration and invasion.

Conclusion

LncRNA SNHG17 is highly expressed in colorectal cancer and promotes the migration and invasion of colorectal cancer cells by inducing epithelial-mesenchymal transition.
