目的

分析血小板膜糖蛋白GPⅡb/Ⅲa表达水平与结直肠癌临床病理参数的相关性。

方法

对河北省人民医院肿瘤95例结直肠癌患者及40例健康志愿者进行外周血血小板计数、流式细胞学检测同时空腹查外周血GPⅡb/Ⅲa的表达水平；以CD31为内皮标志物，采用免疫组织化学方法对95例结直肠癌标本进行微血管染色。

结果

结直肠癌患者血小板的数量明显高于健康志愿者中血小板的数量[(251.61±83.71)×109/L与(184.71±34.48)×109/L(P<0.05)]；血小板的数量与TNM分期相关，其中Ⅰ期数量为(218.81±98.77)×109/L，与健康志愿者差异无统计学意义，Ⅱ期、Ⅲ～Ⅳ期的数量分别为(248.34±70.39)×109/L、(261.05±88.17)×109/L，明显高于健康志愿者(P<0.05)，而血小板水平与年龄、性别、分化程度、CEA水平、淋巴结转移、浸润深度无明显相关性；结直肠癌患者的GPⅡb/Ⅲa的表达水平明显高于健康志愿者[(25.95±16.92)与(4.45±2.49)，(P<0.05)]，与淋巴结转移[(31.90±17.92)与(19.34±13.00)，(P<0.01)]、浸润程度[(28.01±17.22)与(12.95±5.32)，(P<0.02)]及TNM分期密切相关；GPⅡb/Ⅲa表达水平与微血管密度(MVD)(r＝0.632，P<0.01)成正相关。

结论

血小板膜糖蛋白GPⅡb/Ⅲa的表达水平在结直肠癌的发生、发展过程中至关重要，有望成为结直肠癌治疗的新靶标。

Objective

To explore the correlationship between the level of platelet membrane glycoprotein GPⅡb/Ⅲa and clinical pathological characteristics in colorectal cancer(CRC).

Methods

The number of platelets was detected by routine blood test.The level of GPⅡb/Ⅲa was detected by Flow Cytometer.The expression of microvessel density(MVD)in CRC tissue was assessed by Immunohistochemistry.

Results

The number of platelets of CRC patients was significantly higher than that of control group [(251.61±83.71)×109/L vs(184.71±34.48)×109/L, (P<0.05)]. The level of platelet GPⅡb/Ⅲa in CRC patients was significantly higher than that in control group[(25.95±16.92)vs(4.45±2.49), (P<0.05)], and it was correlated with lymph node metastasis [(31.90±17.92)vs(19.34±13.00), (P<0.01)]. The level of platelet GPⅡb/Ⅲa in stage T3-4 was higher than that in stage T1-2[(28.01±17.22)vs(12.95±5.32), (P<0.02)]; and it was correlated with TNM staging(P<0.05).

Conclusion

The level of GPⅡb/Ⅲa may be involved in the development or progression of colorectal cancer.
