Abstract:
Objective Relying on the Institute of Colorectal Cancer of Harbin Medical University, a standardized biobank of colorectal cancer was established in the northeast of China. To explore the collection, preservation of colorectal tumor tissue samples, construction method of biobank, sample data management and quality control of stored samples, which can provide high quality samples of colorectal tumors occurrence, development, metastasis, biomarker research, translational medicine and individualized treatment.
Methods The colorectal cancer tissues, para-carcinoma tissues and distal normal tissues were resected from colorectal cancer surgery at Harbin Medical University since 2013. The samples were stored at -80℃ low temperature freezer for cryopreservation. At the same time, a random DNA and RNA quality test is carried out on the sampled samples.
Results Up to October 2017, a total of 1 828 samples of tumor, para-carcinoma tissues, normal tissue and a small amount of blood samples were collected from 440 patients. In addition, the quality of DNA and RNA detected by random sampling are in accordance with the quality standards of samples, which can meet the needs of scientific experiments.
Conclusion We have initially established a standardized colorectal tumor biobank, established standardized biobank operation and management procedures, improved the efficiency of biological sample utilization, and provided precious biological sample resources for basic and clinical research of colorectal tumors.
