The correlation between SIRT6 expression and pathological features in patients with colon cancer

Abstract:

Objective

To study the correlation between SIRT6 expression and pathological features in patients with colon cancer and its possible mechanism.

Methods

Sixty patients with colon cancer from April 2015 to April 2017 in Department of General Surgery, Fifth Affiliated Hospital of Harbin Medical University were included in this study, all colon cancer tissues and para cancerous normal colon tissues by surgical excision were taken as pathological specimens.The expression levels of SIRT6 protein and HIF-1α were detected by Western blot method and immunohistochemical method.The clinical and pathological characteristics of gender, age, tumor TNM stage and differentiation in 60 patients were collected. The levels of SIRT6 and HIF-1α in colon cancer and paracancerous tissues were recorded. The difference of pathological features of SIRT6 with different expression levels were compared. The relationship between SIRT6 and pathological features and HIF-1α were analyzed.

Results

The protein expressions of SIRT6 and HIF-1α in colon cancer tissues by western blot were (1.87±0.65) and (1.72±0.59), the levels of SIRT6 and HIF-1α in the para cancerous normal colon tissues were (2.34±1.13) and (0.84±0.36). There were significant difference of the expression of SIRT6 and HIF-1α between colon cancer tissues and the para cancerous normal colon tissues (t=2.793, 9.862; P=0.006, 0.000).The positive SIRT6 protein were 13 cases and the negative were 47 cases among the 60 patients.The logistic multivariate analysis showed that tumor staging was the independent factor, which affected the high expression of SIRT6 protein in colon cancer tissues (OR=0.335; P=0.000). The expression of SIRT6 protein was negatively correlated with the level of HIF-1α in colon cancer tissues (r=-0.819, P=0.000).

Conclusion

The expression of SIRT6 protein in colon cancer tissues are closely related to tumor staging.The SIRT6 protein may inhibit the pathological process of colon cancer, which may be related to the regulation of HIF-1α by SIRT6.
