The effect and mechanism of lysophosphatidic acid oil metastasis and anti-apoptosis in colorectal carcinoma

Abstract:

Objective

This study aimed to investigate whether LPA could synergize TGFβ1 to increase epithelial-to-mesenchymal transition(EMT).

Methods

Western blotting was used to examine the EMT markers: E-cadherin and Vimentin.MTT was used to examine the cell proliferation of 5-Fu and LOHP with or without LPA.Western blotting was used to examine the apoptosis markers Bcl-2, Bax and Caspese-3 after LPA treatment.

Results

TGFβ1 and LPA induced the morphology change of colorectal cancer cell line SW480, and combination group LPA/TGFβ1 induced more significant change.Compared with the LPA group, the difference of E-cadherin in blank control group 1%FBS, and TGFβ1 treatment group, the LPA/TGFβ1 co-treatment group were significant, while the difference of Vimentin was not significant.

Conclusion

LPA synergizes with TGF-β to play an important role in EMT.Inhibition of LPA may decrease the invasion and migration of colon cancer cells.
