血浆外泌体HSP90α对预测结直肠癌肝转移的价值探索

目的

探讨结直肠癌患者血浆外泌体HSP90α与结直肠癌肝转移的关系。

方法

应用超速离心法提取27例I~III期原发性结直肠癌患者和27例结直肠癌肝转移患者的血浆外泌体,并应用ELISA法检测其HSP90α表达,并与相同患者血浆游离HSP90α比较。

结果

应用超速离心法能有效提取血浆外泌体,经验证符合外泌体相关特征。结直肠癌患者血浆外泌体HSP90α表达水平与患者肿瘤部位有关(H=6.426,P<0.05)。结直肠癌肝转移患者血浆外泌体HSP90α的表达水平高于原发性结直肠癌患者(Z=2.898,P<0.05),与血浆游离HSP90α表达趋势相同。多因素logistics回归分析显示血浆外泌体HSP90α、血浆游离HSP90α、CEA是诊断结直肠癌肝转移的独立危险因素(P<0.05)。血浆外泌体HSP90α联合CEA具有最高的诊断效能。

结论

血浆外泌体HSP90α可能与结直肠癌肝转移生物学功能相关,可以作为传统标志物的有效补充。

Objective

To explore the relationship between plasma exosome HSP90α and colorectal cancer liver metastasis in patients with colorectal cancer.

Methods

The differential expression levels of plasma free HSP90α and exosome HSP90α extracted by ultracentrifugation method were detected by ELISA in 27 patients with stage I-III primary colorectal cancer (pCRC) and 27 patients with colorectal cancer liver metastasis (CRLM).

Results

Ultracentrifugation method can effectively extract plasma exosomes, which have been verified to meet the relevant characteristics of exosomes. The expression level of plasma exosome HSP90α in CRC patients is related to the degree of tumor site (H=6.426, P<0.05). The expression levels of plasma exosome HSP90α in CRLM group were significantly higher than those in pCRC group (Z=2.898, P<0.05), which is consistent with the expression trend of plasma free HSP90α. Multivariate logistics regression analysis showed that plasma exosomes HSP90α, plasma free HSP90α, and CEA were independent risk factors for the diagnosis of colorectal cancer and liver metastasis (P<0.05). And plasma exosome HSP90α combined with CEA has the highest diagnostic efficiency.

Conclusion

Exosome HSP90α may be related to the biological function of colorectal cancer liver metastasis, and can be used as an effective supplement to traditional markers.
